losing shape of protein is called denaturation, where a folded form of protein is unfolded or misfolded. It can be occured by chemicals, heat or even cells have proteins which can denature different proteins.

After denaturation protein loses its function but yes there are some proteins which are active when misfolded called as Intrinsically disordered proteins.
