This is because when you have a negative exponent like -x to rearrange it to a fraction and change the negative to a positive.
So 4^x would be the denominator and 1 would be the numerator.
So 1 / 4^x = 1 / 16
To achieve 16 from 4 you square it, therefore x = 2
