Hydrophobic signaling molecules ( ligands ) can diffuse through the plasma membrane and bind to internal receptors. Water-soluble ligands are unable to pass freely through the plasma membrane due to their polarity and must bind to an extracellular domain of a cell -surface receptor.

I hope this helped you!<3!
