In which layer is lithification most likely to occur?

Lithification is the process by which sediment turns into hardened rock. There are three ways in which lithification can occur. These processes are called compaction, recrystallization and cementation.

It happens mostly in the subsoil.

Lithification is the process of sedimentation and compaction of soft materials to form rocks. The materials are soft due to the presence of liquids mostly water. Using high pressure, water is removed from the materials and made more compact. Sedimentary rock layers are result of lithification.
