How do the connotations of words affect the theme of a poem?

It would depend on the specific words and the specific poem.

This is sort of like asking "how do the ingredients affect baked goods?" You can't really say unless you know what sort of baked goods you're talking about (bread? cookies?) and which ingredients (what kind of flour? Inclusion of baking powder or not?) etc.
