Macromoleucles such as proteins are too large to pass through the membrane. The macromolecules are loaded into small membrane wrapped like structure called vesicles. A vesicle is formed when the membrane is bulges out or pinches off from the Golgi bodies. After reaching its destination it merges with another membrane to release its macromolecules.
