1. Sample WBC is required to be kept alive for observation:

1. Phase-contrast microscopy: It is a type of light field microscopy that enhances the contrasts of transparent and colourless objects to give a clear view of their edges.
2. Confocal microscopy: Through the use of laser to illuminate different layers of the WBC.

2. Sample WBC can be killed for observation: Transmission electron microscope can be used to view the cross section of the WBC. The various organelle will be visible in a great detail through this technique of microscopy.
